Effects of Phenylhydrazine on Phosphatase Activity in Rat Erythrocytes

Abstract
|
|
|
The activity of alkaline and acid phosphatase was studied in the red blood cells from phenylhydrazine treated rats, during induction of reticulocytosis and recovery period toward normal value.
The activity of alkaline phosphatase was increased significantly in the cytosolic and mitochondrial fractions after phenylhydrazine treatment. Each maxial valus was attained on the 4th day after treatment, which was about 1.7 times higher than the control value for cytosolic isozyme and was about 3 times for mitochondrial isozyme.
In the case of acid phosphatase, the activity was also increased reached a maximal value on day 4, which was about 1.2 times higher than the initial value for cytosolic isozyme and was about 5.5 times mitochondrial isozyme.
Also, we have studied the kinetics of each enzyme (the first-order rate constant for synthesis(Ks) and for degradation(Kd) and the half life(T1/2).
This results suggested that, by phenylhydrazine, mitochondrial enzymes should be more inducible than cytosolic enzymes and the induction of phosphatase should be faster than that of reticulocytosis.
